Cell Staining Buffer
Introduction
Cell Staining Buffer, a buffer optimized for sample preparation of flow cytometry (FCM), can be used in fluorescent antibody dilution and cell sample washes.
This product contains bovine serum albumin (BSA) as carrier protein to reduce non-specific binding of fluorescent antibodies. Experiments showed that addition of BSA can also reduce damage to cells during sample preparation and retain more cells for analysis.
This product contains EDTA as a chelating reagent for metallic ions which promote cell to cell adhesion. Addition of modest amount of EDTA can boost the ratio of singlets in FCM analysis. This product contains preservative to prevent bacterial contamination at a safe dosage to make sure that the impact on cells can be negligible.
Instructions
This product is provided as 1× working buffer. It can be added directly into tubes containing cell pellets to resuspend.
Experimental Procedure
- Single cell suspensions are centrifuged at 300 g for 5 min, discard the supernatant.
- Add desired amount of Cell Staining Buffer with a pipettor to resuspend the cell pellets. For common analysis, 106 cells are resuspended with 100 μL buffer, i.e. with a cell density of 107 cells/mL.
Storage
Store at 2~8°C. Avoid of freezing. Valid for 12 months.
