Overview

MALDI-TOF is a novel soft ionization mass spectrometry technique developed in recent years for organic compounds. It detects samples based on their mass-to-charge ratio (m/z) and measures the molecular weight of sample molecules. It provides accurate measurements for molecular weights under 10,000, but accuracy decreases significantly above this range. Theoretically, the instrument can measure up to 200,000.

Typical sample types include proteins, nucleic acids, peptides, and macromolecules. Based on the results, it is categorized into low resolution and high resolution: low resolution is accurate to one decimal place, while high resolution is accurate to four decimal places.

There are two testing modes: linear mode and reflection mode. Linear mode is the default; please indicate if reflection mode is needed. Each mode is further divided into positive ion and negative ion modes, depending on the molecular structure, as some molecules are more likely to carry a positive charge and others a negative one. Positive ion mode detects positively charged ions, while negative ion mode detects negatively charged ions. By default, the positive ion mode is used.

Common test matrices include CHCA, DHB, and SA, with CHCA as the default. For special testing requirements, please consult us.

Sample Requirement

  • Sample State: Samples can be in powder or liquid form and should ideally be free of salts.
  • Powder Samples: Fine powder, 5-10 mg, which can dissolve or disperse in a suitable solvent. Common solvents include CDCl₃, toluene, acetone, methanol, acetonitrile, trifluoroacetic acid, water, ethyl acetate, ether, and THF.
  • Liquid Samples: 2 mL, with the specific concentration of the liquid sample noted (typically required to be at least 1 mg/mL). Additionally, please specify the buffer composition (generally recommended buffers include Tris or ammonium bicarbonate, with concentrations below 20 mM classified as salts, which are easily ionized; higher concentrations may interfere with results).
