Test Principle

This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human RBP4. Samples (or Standards) and biotinylated detection antibody specific for Human RBP4 are added to the micro ELISA plate wells. Human RBP4 would combine with the specific antibody. Then Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human RBP4, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 ± 2 nm. The OD value is proportional to the concentration of Human RBP4. You can calculate the concentration of Human RBP4 in the samples by comparing the OD of the samples to the standard curve.

Background

Retinol binding protein 4 (RBP4), previously called retinol binding protein (RBP) is a transporter protein. This protein belongs to the lipocalin family and is the specific carrier for retinol (vitamin A alcohol) in the blood [1]. It delivers retinol from the liver stores to the peripheral tissues. In plasma, the RBP-retinol complex interacts with transthyretin which prevents its loss by filtration through the kidney glomeruli. A deficiency of vitamin A, blocks secretion of the binding protein post translationally and results in defective delivery and supply to the epidermal cells.RBP4 seems to be correlated with cardio metabolic markers in inflammatory chronic diseases, including obesity, type 2 diabetes, metabolic syndrome, and cardiovascular diseases (CVDs) [2].

Assay Procedures

step01

1. Add 50μL standard or sample to the wells, immediately add 50μL Biotinylated Detection Ab working solution to each well. Incubate for 90 min at 37°C

step02

2. Aspirate and wash the plate for 3 times

step03

3. Add 100μL HRP conjugate working solution. Incubate for 30 min at 37°C. Aspirate and wash the plate for 5 times

step04

4. Add 90μL Substrate Reagent. Incubate for 15 min at 37°C

step05

5. Add 50μL Stop Solution

6. Read the plate at 450nm immediately. Calculation of the results
