MSE PRO™ Collagenase, Type 7

The MSE PRO™ Crude collagenase preparations contain several isoforms of two different collagenases, mercapto proteases, clostridial enzymes, trypsin-like enzymes and aminopeptidases. The combination of collagen-hydrolyzing and protein-hydrolyzing activities allows for efficient breakdown of the intercellular matrix, resulting in tissue dissociation. One component of the complex is a hydrolase that preferentially degrades the helical region in natural collagen at the Y-Gly bond in the sequence Pro-Y-Gly-Pro, where Y is usually a neutral amino acid.

This cleavage yields a product that is sensitive to further peptidase digestion. Metal chelators such as cysteine, EDTA, and o-phenanthroline inhibit collagenase, but DFP does not. It is also inhibited by α2-macroglobulin, a large plasma glycoprotein. The enzyme requires calcium ions for activation. The specific enzyme profile of each collagenase has been correlated with the tissue from which the cells used for the study were obtained (or the purpose for which the cells were used).
