The MSE PRO™ Most published conventional methods for isolating hepatocytes use crude and partially purified enzyme preparations, including various types of collagenases and other proteases. Recently, better results have been achieved using better characterized collagenase preparations such as our types 1 and 4 (CLS-1, 4). All crude collagenase preparations may contain different batches of contaminating proteases, esterases, and other enzymes, requiring researchers to pre-screen multiple batches of enzymes and/or continually modify separation parameters and protocols. By using the pre-optimized enzyme combinations in this kit, batch-to-batch variation can be minimized and the quality of isolated hepatocytes can be improved. In addition, we tested each batch for use by isolating hepatocytes from adult rats to ensure performance, reliability, and consistent yields of viable cells.
