The MSE PRO™ Our goal in developing the Neonatal Cardiomyocyte Isolation System was to provide researchers with a reliable, convenient, and consistent method for isolating neonatal rat cardiomyocytes. By using purified enzyme preparations rather than crude enzyme preparations, batch-to-batch variation can be minimized. In addition, we have tested the kit in use by isolating cardiomyocytes from neonatal rat hearts to ensure performance, reliability and consistent live cell yields. The kit was formulated in collaboration with Dr. Ronal MacGregor. The method is based on that described by Toraason et al. in Toxicol, 56, 107 (1988), in which minced tissue is incubated with purified trypsin at 2-8°C overnight. As noted by Toraason, this step reduces the hands-on time required to harvest the cells compared to the time required for continuous incubation in warm trypsin or collagenase. The use of purified collagenase rather than crude collagenase maximizes yield and viability.
