KW Parn et al investigate the anti-hypertension effect of epigallocatechin gallate. Thiobarbituric acid reactants (TBARS) level in rat liver was determined using TBARS colorimetric assay kit (E-BC-K298-M).

High dose EGCG increased the concentration of TBARS in rat liver. (*P<0.05)

 

Dilution of Sample

It is recommended to take 2~3 samples with expected large difference to do pre-experiment before formal experiment and dilute the sample according to the result of the pre-experiment and the detection range (2.6-100 μmol/L).

The recommended dilution factor for different samples is as follows (for reference only):

Detection Principle

TBARS and TBA can react under high temperature and acid conditions and then form a pink compound, the concentration of which is linearly related to the concentration of TBARS in the sample. The TBARS concentration can be calculated by measuring the OD values at 530-540 nm.

Sensitivity

The analytical sensitivity of the assay is 0.85 μmol/L. This was determined by adding two standard deviations to the mean O.D. obtained when the zero standard was assayed 20 times, and calculating the corresponding concentration.
