MSE PRO™ Collagenase, Type 4
The MSE PRO™ Collagenase, Type 4, Crude collagenase preparations contain several isoforms of two different collagenases, a mercapto protease, clostridial protease, a trypsin-like enzyme and an aminopeptidase. This combination of collagenolytic and protein hydrolytic activities effectively breaks down the intercellular matrix and is an important component of tissue dissociation. One component of the complex is a hydrolase that preferentially degrades helical regions in natural collagen, preferentially at the Y-Gly bond in the sequence Pro-Y-Gly-Pro, where Y is usually a neutral amino acid. Crude collagenase is inhibited by metal chelators such as cysteine, EDTA or o-phenanthroline, but not by DFP. It is also inhibited by α2-macroglobulin (a large plasma glycoprotein). The enzyme requires calcium ions for activation.
The specific enzyme profile of each collagenase correlates with the organization of the cells used for the study (or the purpose for which the cells were used). And because of the correlation, we have established several types of crude collagenase: type 1, type 2, type 3, and type 4. type 1 crude collagenase has a primitive proportion of collagenase with tyrosinase, clostridial protease, and tryptic activity. type 2 contains higher relative levels of protease activity, especially clostridial protease. type 3 contains the lowest levels of secondary proteases. type 4 tryptic activity is particularly low to limit damage to membrane proteins and receptors. Purified collagenase contains the least amount of secondary protein hydrolyzing activity as well as high collagenase activity. Levels of secondary proteases are similar to Type 1 and Type 2 collagenases.
