MSE PRO™ Ribonuclease T1, Chromatographically Purif., Lyophilized
The MSE PRO™ Ribonuclease T1 is an endonuclease with high specificity for cleaving RNA or deamidated RNA between a 3'-phosphoguanosine residue (or 3'-phosphoinositide) and the 5'-OH residue of an adjacent nucleotide and forming the corresponding 2', 3'-cyclic phosphate intermediate. It cleaves single-stranded RNA and releases oligonucleotides from the 3'-phosphoguanosine terminus. The molecular weight of the enzyme is 11 kDa. The optimum pH is 7.5. The stimulatory effect of histidine and ethylenediaminetetraacetic acid is due to chelation of contaminating inhibitory cations. The enzyme assay is essentially the method of Egami et al, Prog. in Nucleic Acid Res. and Molec. Biol., III, 59 (1964), which is based on the release of acid-soluble oligonucleotides after yeast RNA digestion.
