MSE PRO™ Ribonuclease T2, Recombinant

The MSE PRO™ Aspergillus oryzae ribonuclease T2 is a member of the RNase T2 family of endonucleases found in a wide range of microbial, plant and animal species. Unlike Aspergillus oryzaeRibonuclease T1, a nucleic acid endonuclease that is exclusively guanosine-specific, all RNase T2-like enzymes are essentially non-base-specific. However, RNase T2 endonucleases from different species exhibit slight base preferences. Fungal enzymes, including Pseudomallei RNaseT2, show slight base preferences in the following order: RNase T2 cleaves between the 3'-phosphate residue of one base and the 5'-OH residue of an adjacent nucleotide to form a 2',3'-cyclic phosphate intermediate, which then generates an oligonucleotide with a 3'-phosphate residue. RNase T2 has a molecular weight of 36 kDa and 12-15% of its mass consists of carbohydrates. It has an isoelectric point of 5.0 and is optimally active at pH 4.5. RNase T2 is strongly inhibited by Cu2+, Zn2+ and Hg2+, followed by Ca2+, Mg2+ and heparin. Single nucleotides and RNase T2 digestion products also act as competitive inhibitors. Ethylenediaminetetraacetic acid stimulates activity, especially in the presence of divalent cations.
